This gene encodes a five transmembrane protein that functions as a major regulator of the innate immune response to viral and bacterial infections. The encoded protein is a pattern recognition receptor that detects cytosolic nucleic acids and transmits signals that activate type I interferon responses. The encoded protein has also been shown to play a role in apoptotic signaling by associating with type II major histocompatibility complex. Mutations in this gene are the cause of infantile-onset STING-associated vasculopathy. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Sep 2014] Database links: Entrez Gene: 340061 Human SwissProt: Q86WV6 Human STING由379個氨基酸構成，有數(shù)個跨膜結構域，在多種內皮細胞、上皮細胞中以及造血肝細胞均有表達。STING參與并促進DNA介導的先天免疫反應，即來自病毒和細菌的非CpG雙鏈DNA與cGAS蛋白結合后被催化成第二信使cGAMP；接著二聚化的STING與cGAMP結合，發(fā)生構象變化，通過自噬小體（autophagsome）經(jīng)內質網(wǎng)和高爾基體進一步進入細胞質，期間被泛素化后募集TBK1蛋白，再被磷酸化，之后與IRF3結合；接著IRF3被磷酸化后入核，誘導表達IFNs，細胞因子、T細胞募集因子等進而增強機體的免疫能力，更大限度的殺傷細菌，病毒等病原體。 |
