The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor is expressed by activated, but not by resting, T and B cells. TRAF2 and TRAF5 can interact with this receptor, and mediate the signal transduction that leads to the activation of NF-kappaB. This receptor is a positive regulator of apoptosis, and also has been shown to limit the proliferative potential of autoreactive CD8 effector T cells and protect the body against autoimmunity. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008] Function: Receptor for TNFSF8/CD30L. May play a role in the regulation of cellular growth and transformation of activated lymphoblasts. Regulates gene expression through activation of NF-kappa-B. Subunit: Interacts with TRAF1, TRAF2, TRAF3 and TRAF5. Subcellular Location: Isoform Long: Cell membrane; Single-pass type I membrane protein. Isoform Short: Cytoplasm. Tissue Specificity: Detected in thymus and in activated splenocytes. Post-translational modifications: Phosphorylated on serine and tyrosine residues. 25 ug total protein per lane of various lysates (see on figure) probed with CD30 monoclonal antibody, unconjugated (bsm-54798R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
Paraformaldehyde-fixed, paraffin embedded Human Tonsil; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with CD30 Monoclonal Antibody, Unconjugated(bsm-54798R) at 1:200 overnight at 4°C, followed by conjugation to the bs-0295G-HRP and DAB (C-0010) staining.
